| Abstract: | A factor which determines availability of 02 to tissues is the oxy-hemoglobin dissociation curve position, which is dependent in part on 2,3 DPG concentration. During a saturation dive of three healthy volunteers to pressures equivalent to 650 msw (Atlantis IV) we reported that pressure exposure was accompanied by a small leftward shift (2.3 mmHg) in P-50 of blood obtained and analyzed at depth (Stolp et al., VIII Symposium, 1984), a shift considerably smaller than expected from the work of Kiesow in vitro (UBR, 1974). We wished to test the hypothesis that pressure-induced decrease of P-50 may be accompanied by a compensatory increase in 2,3 DPG, which would tend to normalize P-50. During the dive, blood samples were obtained at various ambient pressures in order to assay the levels of 2,3 DPG (DPG), while subjects were breathing .5 ATA 02' Controls were obtained in air before and after the dive. Blood samples were rapidly fixed with perchloric acid and decompressed. DPG was assayed fluorometrically. Mean values obtained are reported in the Table as uM(DPG)/g Hb. 1 ATA Control Atlantls IV Post-dlve X ± 1 SO 15.9 ± 3.5 13.7 ± 2.6 15.1; (n) (12) (19) (2) We conclude that DPG during pressure exposure was not elevated above 1 ATA values. Furthermore, the 14% reduction in DPG was not sufficient to explain fully the magnitude of P-50 shift previously reported. |
