Abstract: BACKGROUND: Previous reports suggest an involvement of the L-arginine-NO pathway in the pathophysiology of hyperoxia-induced seizures, based on the use of systemic NO synthase (NOS) inhibitors and the NO precursor L-arginine. In order to further evaluate the role of NO we used two agents which modulate NO levels - the NO donor S-nitroso-N-acetylpenicillamine (SNAP), and the novel specific cerebral NOS inhibitor 7-nitroindazole (7NI). METHODS: Male rats implanted with chronic cortical electrodes for continuous EEG monitoring were used. Fifty rats were treated with either 7 NI (50 mg/kg), SNAP (250, 500, and 1000 (g/kg), or their vehicles, before exposure to oxygen or oxygen with 5percent CO2 at 0.5 MPa. The duration of the latent period until the appearance of electrical discharges in the EEG was used as an index of CNS oxygen toxicity. RESULTS: Pretreatment with 7 NI significantly prolonged the latent period to the onset of seizures compared to the vehicle both on exposure to pure oxygen at hyperoxic-hypercapnic mixture (45.6+/-4.4 vs. 26.6+2.8 at 0.5 MPa oxygen, and 34.1+/-0.9 vs. 8.7+/-0.3 min in 0.5 MPa oxygen at 5percent CO2 respectively, P less than 0.05). SNAP significantly shortened the latent period (17.7+/-2.4 vs. 28.6+/-3.5 min, P less than 0.05). CONCLUSIONS: Our results demonstrate a protective effect of the cerebral NOS inhibitor 7NI in hyperoxia-induced seizures, and increased toxicity after treatment with the NO donor SNAP. These findings further confirm the role of NO in the development of hyperoxia-induced seizures.
